Several factors affecting the effect of coagulation:

1. Blood coagulation speed: The blood coagulation mechanism is a process in which a series of coagulation factors are activated one after another, and finally a fibrin clot is formed. In vacuum blood collection tubes containing coagulant, sometimes fibrin precipitated filaments and lumps may appear, which is caused by the non-standardized use of coagulation blood collection tubes. When preparing the vacuum blood collection tube, choosing a coagulant with a fast coagulation speed will cause the fibrin to contract too fast and fragment the fragile red blood cells, causing mild hemolysis. The blood clot is larger than the serum, so the blood clot is on the top and the blood clot is on the bottom. The upper end of the blood cell is still in contact, and the cells can still use the nutrients in the serum to decrease the blood glucose measurement value, the lactate dehydrogenase, and the serum potassium measurement value. Therefore, the upper layer is serum, the middle layer is separation gel, and the lower layer is blood clots, so the various components in the serum maintain physiological levels.



2. Blood coagulation temperature: The natural coagulation of blood is related to temperature. The blood can coagulate in a glass test tube in a 37° water bath for 30 minutes. However, it should be pointed out that if the blood and the coagulant are not sufficiently mixed after blood collection or the blood is not completely coagulated and centrifuged, it is easy to form fibrin jelly-like coagulation or fibrin filaments whose specific gravity is smaller than that of blood clots, so they remain The serum layer is partially attached to the separation gel. If it is directly connected to the machine at this time, it can cause blockage of the automatic analysis blood sampling needle.



3. The amount of coagulant: the relative amount of coagulant needed to make the blood achieve the best coagulation effect. When the blood collection tube company prepared the coagulant blood collection tube, the coagulant spray was insufficient or the prepared water-soluble coagulant was not used up on the same day, and the coagulant was continued to be used the next day, which resulted in the decrease of the coagulant effectiveness. The fibrinogen in the blood is gradually transformed into insoluble fibrin under the action of the coagulant. If the coagulation dose is insufficient or ineffective and the clotting time is prolonged, centrifugation can lead to the precipitation of fibrin.



4. Whether the operation is standard: The general fibrin precipitation has the following reasons: use the coagulation blood collection tube or the separating gel to promote the coagulation blood collection tube. To make the coagulant evenly distributed in the blood, the coagulant must be slightly inverted and mixed 4 to 5 times. The center of the blood collection tube coagulates at the same time as the surrounding blood. If the blood is not completely coagulated and then centrifuged, it can cause the precipitation of fibrin. The upper part of the serum appears jelly-like and fragmented, which is mixed with the blood. Fibrin filaments are caused by the fact that fibrin has not yet fully contracted. When preparing coagulant vacuum blood collection tubes, coagulant should be sprayed quantitatively and dried under the condition of less than 45°C; if it is higher than 50°C, the effectiveness of coagulant may decrease. The coagulant prepared with distilled water or with absolute ethanol must have standardized quality management, and the coagulant shall be sprayed quantitatively. The blood of dialysis patients must be separated from the serum with a coagulation tube containing neutralizing UFH in order to correctly separate high-quality serum samples.
