The improvement of today's living standards is closely related to the advancement of medical testing technology and the development of medical testing technology in the field of clinical testing. In serology, immunology and other tests, most specimens need to be checked for blood. Blood anticoagulants include separation gel/EDTA/UFH/sodium citrate. This article describes the following steps of anticoagulant in the extraction and separation of hemoglobin. Gel chromatography is used here.

 

Wash with RBC to remove the protein. After collecting the blood, separate the red blood cells in a short time at a low speed, and then use a plastic pipette to suck out the upper transparent yellow plasma, pour the lower red blood cell liquid in the beaker, and then add 5 times the volume of normal saline. Stir slowly for *0 minutes, and repeat the washing three times in a low-speed centrifuge for a short time until there is no yellow in the supernatant, indicating that the red blood cells have been washed. With the release of hemoglobin in the presence of distilled water , red blood cells split to release hemoglobin. The centrifugal stirring is divided into four layers in the test tube.

 

The first layer is a transparent colorless layer, the second layer is a thin white solid precipitate of fat-soluble substances, the third layer is a transparent red liquid is an aqueous solution of hemoglobin, and the fourth layer is a deep red precipitate of other impurities. Filter the liquid in the test tube with filter paper to remove the soluble sediment layer. After standing for a while in the separatory funnel, separate the red transparent liquid from the lower layer.

 

Put 1mL of hemoglobin solution into a dialysis bag, and then put it into a phosphoric acid solution containing **0mL of a substance with a concentration of *0mmol/L for *2h dialysis. Dialysis can remove lower molecular weight impurities from the sample or can be used in place of sample buffer.
