Overview
The Aspergillus Galactomannan Detection Kit is an immunoenzymatic indirect competitive microplate assay for the detection of Aspergillus galactomannan antigen in human serum samples. The assay is used to provide rapid diagnosis reference for the clinicians in identifying Invasive Aspergillosis (IA) early in the disease process.

Principle of the Test
This product is an ELISA indirect competition assay for the detection of Aspergillus galactomannan antigen using specific antibodies. Put the pre-treated human serum sample and the anti-galactomannan antibodies into the wells of the microplate which have been coated with the Aspergillus galactomannan. After the wells are incubated and washed, the enzyme labeled antibodies are added. Following a second time incubating and washing the wells, TMB substrate solution is added to form a color reaction. The absorbance is determined with a microplate reader at **0nm wavelength. The absorbance value has a negative correlation with the galactomannan content. The galactomannan concentration of the sample serum can be calculated from the standard curve, hence the Aspergillus galactomannan quantitative detection can be performed.


Clinical Significance
The incidence of invasive fungal infection (IFI) by both primary and opportunistic pathogens,has been steadily increasing among hematological malignancy and AIDS patients and the patients receiving aggressive chemotherapy for cancer, bone marrow and organ transplantation or advanced critical care. The difficulty in diagnosis of these infections are associated with significant mortality and morbidity. Since the traditional methods for the fungal infection detection such as blood culture and histopathological diagnosis are either less sensitive or not feasible in the critically ill patients, developemnt of untraditional method for the IFI diagnosis is of great importance.
Among the new diagnostic techniques is the assay for the serum (**3)-β-d -glucan (BG) detection. Positive in the BG test has been recognized as a very important microbiological evidence in the clinical diagnostic criteria for invasive fungal infection, which can achieve early diagnosis for IFI and dynamic monitoring among patients with risks of IFI. What is more, BG test results provide basis to the use of medicine and pharmacodynamics evaluation in clinical treatment.

Samples Collection and Storage
Collect venous blood samples 4ml with sterile non-pyrogenic vacuum tubes (red tops). The serum is then separated from the blood by centrifuge (***0 rmp/min for ****5 minutes). Detect within 2 hours.
Store the kit at **8℃ in the dark. Do not freeze any kit component. The kit can be kept for 1 year before opening. Once opened the kit is stable for 4 weeks. Avoid contamination during sample collection, transportation, and preservation. Serum samples should be stored at **8℃or frozen at **0℃ or colder within *4 hours after collection. If the sample could not be detected in time, it should be stored below **0 ℃ to avoid sample contamination and deterioration. The serum samples should avoid repeated freezing and thawing.

Because a fungal infection is a dynamic process repeat testing, typically **3 times per week, improves sensitivity.
